角类动物药特征肽段的寻找研究进展摘 要角类动物药作为一类较为名贵的中药材,特别是羚羊角更是价格奇高。
此类药材具有活性强、疗效显著、副作用小等特点,在中医药领域使用广泛。
但由于资源稀缺,且用药多为粉末,角磅丝等,难以通过外观鉴别,造成市场混乱、伪混品泛滥等情况。
角类药材的专属性鉴定方法研究起步较晚,目前主要的鉴定方法包括显微鉴别、薄层鉴别、氨基酸鉴别、DNA条形码鉴别、特征肽段鉴别等。
其中DNA条形码鉴别、特征肽段鉴别为近年发展起来的新兴鉴定方法。
角类药材的成分以角蛋白为主,而每种生物都有专属性的肽段,寻求一种针对物种角蛋白特征肽段的鉴定方法,此方法在高通量质谱的基础上,规避统计学的分析方法,而采用数学集合的方法来寻找专属性肽段,构建正品的肽段集合与伪品的肽段集合,通过两者相交,取完全属于正品集合,同时完全不属于伪品集合的肽段作为专属性肽段的候选肽段,即回避概率性的分析,而采用全或无的分析思路寻找专属性肽段。
准确率高,重复性好,将对建立角类动物药质量标准,规范角类中药市场具有积极的意义。

However, due to the scarcity of resources, and the use of drugs are mostly powder, shreds, etc., it is difficult to identify through the appearance, resulting in market confusion, the proliferation of fake mixed products. The research on the specific identification method of keratinous medicinal materials started late. At present, the main identification methods include microscopic identification, thin layer identification, amino acid identification, DNA barcode identification, and characteristic peptide identification. Among them, DNA barcode identification and characteristic peptide identification are emerging identification methods developed in recent years. The constituents of keratin are mainly keratin, and each organism has a specific peptide. It seeks a method for identifying the keratin-characteristic peptides. This method avoids statistics based on high-throughput mass spectrometry. The analytical method of learning, and the method of mathematical collection is used to find the specific peptides, and the collection of the authentic peptides and the peptides of the fakes are constructed. By the intersection of the two, the products are completely belong to the genuine collection, and at the same time, they are not belonging to the fake collection. The peptide segment is used as a candidate peptide for the specific peptide segment, that is, the avoidance probability analysis, and the 'all' or 'none' analysis method is used to find the specific peptide. 动物药作为动物的器官、组织或代谢产物,其来源、组成、成分与植物药大相径庭,动物药研究是中药研究领域特有的分支,是具有独特体系的学科,因此采用目前相对较成熟的植物药研究思路去探索动物药的道路是很难走通的。
